Mouse Monoclonal Antibody

This 9E2 monoclonal antibody reacts with human CD335, which is also known as NKp46. Expressed on NK cells, CD335 is a member of the natural cytotoxicity receptor family that also includes NKp30 and NKp44. This type I transmembrane glycoprotein is involved in activating and expanding resting NK cells following interaction with dendritic cells to eliminate tumor and virus-infected (e.g., influenza) cells. CD335 also interacts with pancreatic beta cells during the development of type 1 diabetes. The 9E2 antibody has been reported to induce cytotoxicity. Applications Reported: This 9E2 antibody has been reported for use in flow cytometric analysis. Applications Tested: This 9E2 antibody has been pre-titrated and tested by flow cytometric analysis of normal human periheral blood cells. This can be used at 5 μL (0.125 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. NovaFluor dyes are not compatible with DNA intercalating viability dyes. Do not use viability dyes such as propidium iodide, 7-actinomycin D (7-AAD) and DAPI. Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for use with NovaFluor dyes. Use 5 μL of CellBlox Blocking Buffer per stained cell sample containing 10^3 to 10^8 cells. NKp46 (CD335, NCR1, ) is a cytotoxicity-activating receptor that may contribute to the increased efficiency of activated natural killer (NK) cells to mediate tumor cell lysis. NKp46 consists of two Ig-like domains assembled to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
