Description: This 2SX21R monoclonal antibody reacts with human CD360 (Interleukin-21 Receptor, IL-21R). IL-21R forms a heterodimeric receptor complex with the common gamma chain to form a functional receptor for IL-21. IL-21 is a class I cytokine of the common gamma chain family of cytokines and signals through the IL-21R complex to activate the JAK/STAT (Janus Kinase/Signal Transducers and Activators of Transcription) signaling pathway. Activation of the JAK/STAT pathway by IL-21 promotes transcription of genes responsive to STAT1 and STAT3. IL-21 is mainly produced by CD4+ T cells and mediates the proliferation, differentiation, and effector functions of cells expressing IL-21R such as B cells, T cells, NK cells, dendritic cells and other populations of myeloid cells. IL-21 and its receptor are implicated in the pathogenesis of chronic viral infections, autoimmunity, allergy, organ transplantation, and cancer. Applications Reported: This 2SX21R antibody has been reported for use in flow cytometric analysis. Applications Tested: This 2SX21R antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 μL (0.5 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. The protein encoded by this gene is a cytokine receptor for interleukin 21 (IL21). It belongs to the type I cytokine receptors, and has been shown to form a heterodimeric receptor complex with the common gamma-chain, a receptor subunit also shared by the receptors for interleukin 2 (IL2) and interleukin 5 (IL5). This receptor transduces the growth promoting signal of IL21, and is important for the proliferation and differentiation of T cells, B cells, and natural killer (NK) cells. The ligand binding of this receptor leads to the activation of multiple downstream signaling molecules, including JAK1, JAK3, STAT1, and STAT3. Knockout studies of a similar gene in mouse suggest a role for this gene in regulating immunoglobulin production. Three alternatively spliced transcript variants encoding the same protein have been described.
