Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous IL21

  • Recognizes human IL21
  • IL21 (Interleukin 21) is a cytokine produced predominantly by cluster of differentiation 4 (CD4+) T-cells and natural killer T-cells
    • IL21 receptor (IL21R) is a class I cytokine receptor, requiring dimerization with indispensable common gamma chain subunit to bind IL21
    • IL21 plays role in innate and adaptive immune responses by inducing differentiation, proliferation and activity of multiple target cells (macrophages, natural killer cells, B cells, cytotoxic T cells)
    • Mediates apoptosis in B-CLL cells through up-regulation of BIM, enhances both direct and antibody-dependent cellular cytotoxicity in cells

Process:

  • Antibody specific for IL21 pre-coated onto microwells
  • IL21 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for IL21 is added to detect captured IL21 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
