Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous LGALS9/Galectin-9

  • Recognizes human LGALS9/Galectin-9 protein
  • Galectins (S-type lectins) are a family of β-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions
    • Galectin-9 (LGALS9) is a tendem repeat-type member of galectin family
    • Three isoforms (galectin-9L, galectin-9M, galectin-9S)
    • Galectin-9 is expressed in tissues including lymph nodes and spleen
    • Overexpressed in Hodgkin disease tissue
    • Involved in chemoattraction, apoptosis and regulation of cell differentiation, has anti-allergic effects
    • Ligand for Tim-3 through which galectin-9 can induce Th1 death

Process:

  • Antibody specific for LGALS9/Galectin-9 pre-coated onto microwells
  • LGALS9/Galectin-9 protein in samples is captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for LGALS9/Galectin-9 protein is added to detect captured LGALS9/Galectin-9 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
