Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous PG II

  • Recognizes human PG II
  • Pepsinogen I and II are precursors of pepsin, preproduced by gastric mucosa and released into gastric lumen and peripheral circulation
    • Advanced inflammation of gastric mucosa and its progression toward atrophic gastritis in corpus is associated with a change in serum biomarkers pepsinogen I and II
    • PG II (Pepsinogen II, progastricsin, PGC, pepsinogen C) is an aspartic protease expressed in gastric chief cells and novel marker of Type 2 cells with advantages over current markers used to identify Type II cells in developing lung
      • Involved in proteolysis and peptidolysis

Process:

  • Antibody specific for PG II pre-coated onto microwells
  • PG II protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for PG II is added to detect captured PG II protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
