Integrin subunit alpha V (ITGAV) and integrin subunit beta 6 (ITGB6) are essential components of the integrin family, which play crucial roles in cell adhesion, signaling, and tissue homeostasis. The ITGAV gene is located on chromosome 2q31, encoding the alpha-V subunit, which pairs with various beta subunits, including ITGB6, to form integrin alpha V-containing heterodimers such as alpha V beta 6. These integrins are pivotal in processes like development, immune regulation, and cancer progression. Structurally, integrins are transmembrane heterodimers comprising alpha and beta subunits that bind to extracellular matrix (ECM) proteins, mediating cell-ECM interactions. The ITGB6 gene, located on chromosome 2q24.2, encodes the beta 6 subunit, which specifically dimerizes with the alpha V subunit to form the alpha V beta 6 integrin. This heterodimer is primarily expressed in epithelial tissues and is involved in processes such as epithelial-to-mesenchymal transition (EMT), wound healing, and fibrosis. Both genes are implicated in various diseases; for example, ITGAV variants are associated with immune dysregulation and brain disorders, while ITGB6 is upregulated in cancer and plays a role in tumor progression and fibrosis.
