Description
NucleoSpin™ Blood QuickPure binding plates are to be used in a centrifuge with deep buckets allowing processing of NucleoSpin™ 96 well plates stacked on 96 well blocks. The method is based on a specially treated silica membrane which speeds up the procedure by a combined washing/drying step. The number of washing and drying steps is reduced from 3 to 1. Therefore the hands-on time is minimised. Lysis is achieved by incubation of whole blood in a solution containing large amounts of chaotropic ions in the presence of proteinase K. Appropriate conditions for binding of DNA to the silica membrane of the NucleoSpin™ Blood QuickPure binding strips are created by addition of ethanol to the lysate. Pure genomic DNA is finally eluted under low ionic strength conditions in a slightly alkaline elution buffer and can be used directly for PCR, Southern blotting or any kind of enzymatic reaction. The kit allows isolation of 4 μg to 6 μg of pure genomic DNA from 200 μL whole blood with an A260/280 ratio between 1.60 and 1.90 and a typical concentration of 40 to 60 ng/μL.Kit components: NucleoSpin™ Blood QuickPure binding plates, buffers, proteinase K, accessories.
