Description

  • Strep-Tactin™ TACS Agarose contains agarose macrobeads coated with the streptavidin variant Strep-Tactin™.
  • A special crosslinking permits the use for cell isolation and depletion approaches.
  • It was developed for column-based Traceless Affinity Cell Selection (Fab-TACS™/Nano-TACS™).
  • In this approach, Fab fragments fused to a Twin-Strep-tag™ (Fab-Streps) or nanobodies (Nano-Streps) are bound to Strep-Tactin™ on the agarose matrix.
  • After specifically capturing cells, Fab-/Nano-Streps are released from the Strep-Tactin™ Agarose by biotin addition.
  • In this way, the eluted cells are ready for further downstream applications.
  • The same principle is applicable for exosome isolation (Fab-TACS™ for exosomes)
  • Alternatively to Fab-Streps/Nano-Streps, it is possible to bind Twin-Strep-tagged, desthiobiotinylated or biotinylated antibodies (irreversible binding) to this resin.
  • This makes Strep-Tactin™ TACS Agarose also suitable for negative cell selection or depletion approaches.
  • The suspension can be used for packing of gravity flow columns since the resin is optimized for column affinity chromatography.
  • To pack a column with 1 ml resin bed volume, 2 ml 50% suspension are necessary.
