Description
This antibody recognizes multiple isoforms of XAF-1. In Western blot applications, a doublet is detected at ~30kDa. A suggested positive control is human spleen tissue lysate. PA5-20090 can be used with blocking peptide PEP-0208.
The inhibitors of apoptosis (IAP) proteins prevent cell death both in vitro and in vivo by virtue of inhibition of distinct caspases. The X-linked Inhibitor of Apoptosis, XIAP, is a key member of the IAP family of proteins. The caspase inhibiting activity of XIAP is negatively regulated by at least two XIAP-interacting proteins, XIAP-associated factor 1 (XAF1) and Smac/DIABLO. XAF1 binds to XIAP and antagonizes caspase inhibitory activities of XIAP. Expression of XAF1 triggers a redistribution of XIAP from the cytosol to the nucleus. XAF1 is ubiquitously expressed in normal tissues, but is present at low or undetectable levels in many different cancer cell lines.
