CD226 (DNAM-1) Monoclonal Antibody (11A8.7.4), PE-Cyanine7, eBioscience™, Invitrogen™
Mouse Monoclonal Antibody
Brand: Invitrogen 25-2269-41
Description
The monoclonal antibody 11A8.7.4 reacts to DNAM-1 also known as CD226. DNAM 1 is a 65 kD receptor expressed by peripheral blood T lymphocytes that is involved in intercellular adhesion, lymphocyte signaling, cytotoxicity and lymphokine secretion mediated by cytotoxic T lymphocytes. DNAM 1 is broadly expressed on T cells, NK cells, platelets, monocytes and a subset of B cells. DNAM-1 is also expressed by a subset of CD3 positive thymocytes. The ligands for CD226 are the poliovirus receptor (PVR, CD155) and Poliovirus Related Receptor (PPR2, Nectin-2, CD112). CD226 is involved in cytolytic activity by interacting with integrins and activating protein tyrosine kinase Fyn. The 11A8.7.4 antibody has been shown to increase T and NK cell-mediated cytotoxicity. Applications Reported: This 11A8.7.4 antibody has been reported for use in flow cytometric analysis. Applications Tested: This 11A8.7.4 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 μL (0.25 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light. Fixation: Samples can be stored in IC Fixation Buffer (Product No.
00-8222) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product No. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. Excitation: 488-561 nm; Emission: 775 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser DNAM 1 is a receptor expressed by peripheral blood T lymphocytes that is involved in intercellular adhesion, lymphocyte signaling, cytotoxicity and lymphokine secretion mediated by cytotoxic T lymphocytes. DNAM 1 is broadly expressed on T cells, NK cells, platelets, monocytes and a subset of B cells. DNAM-1 is also expressed by a subset of CD3 positive thymocytes.
