The Premelanosome protein (PMEL) gene, also known as PMEL17, plays a critical role in the formation of pigment cells. It is located on chromosome 12 and codes for a protein critical for melanosome biogenesis, the organelles responsible for melanin production. PMEL forms amyloid fibrils, which serve as a scaffold for melanin deposition, thus influencing pigmentation. The structure includes several domains, particularly a repeat (RPT) domain that governs the formation of fibrillar sheets. Mutations in PMEL can lead to pigmentation disorders and have been associated with conditions such as pigmentary glaucoma. Additionally, its expression and functionality are crucial in various species and can influence pigmentation patterns.
